ABSTRACT.- Silva S.P., Mota R.A., Faria E.B., Casseb A.R., Casseb L.M.N. & Dias H.L.T. 2013. [Comparison of indirect ELISA and indirect immunofluorescence in detection of antibodies to Neospora caninum in buffaloes (Bubalus bubalis).] E-mail: spatroca@yahoo.com.br To compare two serologic tests for detection of antibodies against Neospora caninum in sera from buffaloes, samples were collected from 288 buffaloes of 2 to 10 years of age. To identify the presence of IgG, anti-N. caninum was used for the indirect immunofluorescence assay (IFAT), with title 200 as the cutoff, and for the immunoenzymatic test (indirect ELISA), considering as positive samples with ratio S/P ≥0.5. There were 153 (53.12%) animals positive for N. caninum by IFAT, whilst 50 (17.36%) animals were reactive in ELISA. The presence of antibodies anti-N. caninum demonstrates that the parasite is circulating among buffaloes raised in Pará State, Brazil. ELISA and IFAT tests could be used to diagnose immunoglobulins against this agent. However there was a weak correlation (Kappa = 0.36) between both tests, considering the IFAT as the gold standard.

ABSTRACT.- Souza M.E., Porto W.J.N., Albuquerque P.P.F., Souza Neto O.L., Faria E.B., Pinheiro Júnior J.W. & Mota R.A. 2012. Seroprevalence and risk factors associated with infection by Neospora caninum of dairy cattle in the state of Alagoas, Brazil. Pesquisa Veterinária Brasileira 32(10):1009-1013. Departamento de Medicina Veterinária, Universidade Federal Rural de Pernambuco, Rua Dom Manoel de Medeiros s/n, Recife, PE 52171-900, Brazil. E-mail: rinaldo.mota@hotmail.com The objective of this study was to investigate the prevalence of anti-Neospora caninum antibodies in cattle from milk producing farms of the microregion of Batalha, state of Alagoas, Brazil, as well as to identify the risk factors associated with the infection. Blood samples were collected from 1,004 cattle of 17 farms for the serological investigation regarding the presence of anti-N. caninum antibodies by the Indirect Immunofluorescence Reaction Technique (IMRT). From the total amount of samples analyzed, 77/1,004 (7.67%) were positive and 927/1,004 (92.33%) were negative. The logistical regression identified that cattle from farms without consortium breeding have an infection risk 6.33 (p<0.001; C.I. 2.89-13.10) times higher than cattle from farms with that type of breeding. Cattle from farms where the aborted fetuses are not adequately buried have an infection risk 3.04 (p<0.001; C.I. 1.64-5.63) times higher than cattle from farms with adequate destination of these fetuses. Infection by N. caninum occurs in cattle of the investigated region. The factors identified in our study can be used as risk indicators, so that control measures could be implemented to avoid infection by N. caninum in the herds of this region.

ABSTRACT.- Valença R.M.B., Mota R.A., Anderlini G.A., Faria E.B., Cavalcanti E.F.S.T.F., Albuquerque P.P.F., Neto O.L.S. & Guerra M.M.P. 2011. [Prevalence and risk factors associated with infection by Toxoplasma gondii on commercial swine farms in the state of Alagoas, Brazil.] E-mail: rmbvalenca@ig.com.br This study was aimed to determine the prevalence and to identify risk factors associated with infection by Toxoplasma gondii in pigs from commercial swine farms in the state of Alagoas, Brazil. To compose the sample size of prevalence 342 pigs were used, with 312 sows and 30 boars, from of seven swine farms and distributed in five districts of the state of Alagoas. The serological examination for the detection of anti-Toxoplasma gondii antibodies was achieved by Indirect Immunofluorescence (IFAT), using anti-pig IgG-conjugated to fluorescein isothiocyanate. The analysis of risk factors were performed by the application of questionnaires consisting of objective questions relating to the designer, the general characteristics of the property, the production, reproductive and health management. The prevalence was of 26,9% (92/342) of seropositive pigs. The associated factor was the boars introduction on farms in the last five years (p=0,014; OR=1,83; CI=1,13-2,96). It is concluded that infection with Toxoplasma gondii is widespread in commercial pig farms in the state of Alagoas, Brazil. The results suggest the control of cat population, the performance of cleaning and sanitation practices and laboratory testing to detect of infection by T. gondii in animals to be placed on the farms as measures to reduce the infection rates on region studied.

Laboratório de Doenças Infecto-Contagiosas dos Animais Domésticos, Departamento de Medicina Veterinária, Universidade Federal Rural de Pernambuco, Rua Dom Manoel de Medeiros s/n, Dois Irmãos, Recife, PE 52171-900, Brazil. E-mail: rinaldo.mota@hotmail.com The aim of this paper was to study the Toxoplasma gondii shedding in the semen of naturally infected rams. Sixty-five rams were initially submitted to anti-T. gondii antibody detection by indirect immunofluorescence (IIF). Serologically positive rams were then submitted to semen collection for T. gondii DNA detection. In the serology, 6/65 (9.2%) rams were positive, while in the nested PCR of semen there were 4/6 (66.6%) positive rams. It can be concluded that detection of the proliferative form of T. gondii in semen of naturally infected rams by the nested PCR technique reinforces the need to investigate possible horizontal transmission of this parasite via semen in sheep.

#5 - Occurrence of IgG antibodies anti-Neospora caninum and Toxoplasma gondii in female water buffaloes (Bubalus bubalis) raised in the Brazilian state of Pará, 30(5):443-446

Abstract in English:

ABSTRACT.- Silva S.P., Mota R.A., Faria E.B., Fernandes E.F.T.S., Neto O.L.S., Albuquerque P.P.F. & Dias H.L.T. 2010 [Occurrence of IgG antibodies anti-Neospora caninum and Toxoplasma gondii in female water buffaloes (Bubalus bubalis) raised in the Brazilian state of Pará.] Anticorpos IgG anti-Neospora caninum e Toxoplasma gondii em búfalas (Bubalus bubalis) criadas no estado do Pará. Pesquisa Veterinária Brasileira 30(5):443-446. Laboratório de Investigação e Diagnóstico de Enfermidades Animais, Universidade Federal do Pará, Belém, PA, Brazil. E-mail: spatroca@yahoo.com.br To investigate the occurrence of antibodies IgG anti-Neospora caninum and Toxoplasma gondii, blood samples were collected from 374 adult female water buffaloes originated from 14 production units located in 13 counties of the state of Pará. The sera were subjected to indirect fluorescent antibody (IFA), using titers of 200 and 64 as IFA cut off points for N. caninum and T. gondii, respectively. Hundred and fifty three (40.9%) buffaloes were sero-positive for N. caninum, with 100% of the properties being sources of infection. Four buffaloes (1.1%) in four farms were seropositive for T. gondii, and one buffalo cow (0.27%) was seropositive for the two parasites. The presence of antibodies indicates that these protozoan parasites are circulating among buffaloes from the properties studied, representing a source of infection to other animals, as well as a possible cause of reproductive disorders in this species.

ABSTRACT.- Medeiros E.S., Santos M.V., Pinheiro Jr J.W., Faria E.B., Wanderley G.G., Teles J.A.A. & Mota R.A. 2009. [In vitro evaluation of the efficacy of commercial disinfectants used in pre and post-dipping against Staphylococcus spp. isolated from bovine mastitis.] E-mail: rinaldo.mota@hotmail.com The objective of this investigation was to evaluate the in vitro sensibility of Staphylococcus spp. to several commercially available disinfectants used for pre and post dipping. A total of 60 isolates of Staphylococcus spp., identified as Staphylococcus aureus (50) and Positive coagulase Staphylococcus (10) were obtained from the mammary glands of dairy cows with subclinical mastitis in the regions of Metropolitan Recife, the Agreste and the Zona da Mata of the State of Pernambuco, Brazil. As active ingredients were used a chlorine base (25%), iodine (0.6%), chlorhexidine (2.0%), quaternary ammonium (4.0%), and lactic acid (2.0%) at four specific intervals (15", 30", 60", and 300"). One hundred percent of S. aureus was found to be sensitive to iodine, 93.3% to chlorhexidine, 80% to ammonia, 35.6% to lactic acid, and 97.8% were resistant to chlorine at a 60-minute interval. With respect to the Positive coagulase Staphylococcus (SCP), 100.0% of the isolates were sensitive to iodine, 81.8% to quaternary ammonium, 99.9% to lactic acid, 72.7% to chlorhexidine, and 100% was resistant to chlorine at an interval of 60 minutes. It can be concluded that the highest disinfectant activity in vitro was found to be with iodine and chlorhexidine for S. aureus, and with iodine and lactic acid for SCP. A further conclusion was that it is important to undertake a periodic evaluation of the disinfectants used on the dairy properties in the regions studied, given the variety of sensibilities and resistance to disinfectants used, which may prejudice the control of bovine mastitis caused by Staphylococcus spp.
